CK20 is a 46 kDa cyto keratin that shows positive staining in normal gastrointestinal epithelium, Merkelcells, and urothelium. It is mainly used for the diagnosis of gastrointestinal adenocarcinoma, Merkel cellcarcinoma, and ovarian mucinous tumors. CK20 is often used in combination with CK7 to determine the primary site of cancer.
CK19 (Cyto keratin 19) is present in normal epithelium and various tumors of epithelial origin, especially insimple epithelium and mesothelium. It is often used for the diagnosis of adenocarcinoma. Hepatocytes donot express CK19, so it can be used for the differential diagnosis of liver cancer and metastatic adenocarcinoma.

Cytokeratin 10 (CK10) has a molecular weight of 56.5 kDa and mainly labels the suprabasal and granular layers of the epithelium. Cytokeratin 13 (CK13) is a marker of undifferentiated non-keratin izing stratified squamous epithelium and is highly expressed in well-differentiated tumor tissues. The expression of CK10 and CK13 often accompanies the occurrence, differentiation, and morphological changes of epithelial cells,and can be used for the study of the occurrence, development, metastasis, and prognosis of tumors derived from stratified squamous epithelium.
Research literature shows that CK8/18 is expressed in simple or glandular epithelium in tumors or normaltissues, such as thyroid, breast, gastrointestinal tract, and respiratory epithelium. It is expressed in adenocarcinoma and most non-keratin izing squamous cell carcinomas, but not in keratin izing squamous cell carcinomas. Therefore, this antibody is usually used in combination with CK5/6 for the differential diagnosis of adenocarcinoma and squamous cell carcinoma.
Cytokeratin 8 (CK8) is also known as tissue polypeptide antigen (TPA). It is one of the earliest keratins expressed during embryogenesis and persists in adult tissues. CK8 and CK18 are major components of all simple epithelia but are not expressed in stratified squamous epithelium. It has been reported that CK8 is expressed in tissues of patients with adenocarcinoma and can also be detected in the serum of such patients.
CK7 is a simple epithelial keratin mainly expressed in glandular epithelium and transitional epithelium. The combined use of CK7 and CK20 is an important tool for determining the primary site of adenocarcinoma. CK7+/CK20- suggests lung adenocarcinoma, breast cancer, etc.; CK7-/CK20+ suggests colorectal cancer.
